Activating Compound

EC Number Activating Compound Comment Organism Structure
2.7.7.86 Plasmodium falciparum genomic DNA activates cGAS in vivo in THP-1 cells, no effect in enzyme-deficient cGAS-/- THP-1 cells. 2',3'-cGAMP is produced in Pf gDNA-transfected THP-1 cells but not in control THP-1 cells Homo sapiens

General Information

EC Number General Information Comment Organism
2.7.7.86 physiological function Plasmodium falciparum (Pf, strain 3D7) genomic DNA (gDNA) delivered to the cytosol of human monocytes (THP-1 cells) binds and activates the cyclic GMP-AMP synthase (cGAS). Activated cGAS synthesizes 2',3'-cGAMP, which acts as a second messenger for STING activation and triggers STING/TBK1/IRF3 activation, resulting in type I interferon (IFN) production in human cells. This induction of type I IFN is independent of IFI16. cGAS is an important cytosolic sensor of Plasmodium falciparum genomic DNA (Pf gDNA) and reveal the role of the cGAS/STING pathway in the induction of type I IFN in response to malaria parasites Homo sapiens
